Effect of Non-Ionic Detergent Tween 80 on Drug-Induced Hemolysis

Abstract
|
|
|
Non-ionic detergent Tween 80 was found to be a potent inhibitor of the hemolytic action of hematin and other detergents such as sodium dodecyl sulfate, deoxycholate, Triton X-100, and saponin. In contrast, the p-hydroxymercuribenzoate-induced hemolysis was markedly enhanced by Tween 80.
In addition, Tween 80 inhibited the binding of hematin to the human erythrocyte membrane preparation and decreased the osmotic fragility of human erythrocytes. It also increased the erythrocyte sedimentation rate, and decreased the sulfhydryl content of the erythrocyte membranes which is reactive with 5,5¡¯ - dithiobis(2-nitrobenzoic acid). However, when Tween 80 was washed off from the Tween 80-pretreated erythrocytes with isotonic NaCl solution, the subsequent p-hydroxymercuribenzoate-induced hemolysis was not enhanced.
These results suggest that Tween 80 reversibly binds to the erythrocyte membrane in preference to the other drugs, and prevents the binding of them to the membranes. These results also suggest that Tween 80 has little deleterious effect on the erythrocyte membrane.
